Zhigang's work has been published in Chem. We reported phorbiplatin, the first small-molecule Pt(IV) prodrug that can be efficiently and controllably activated by red light. The prodrug utilizes a unique photoreduction mechanism to release oxaliplatin, resulting in significantly enhanced antitumor activity both in vitro and in vivo.

Dr. Zhu’s work on a monofunctional platinum-based anticancer agent, pyriplatin, has been recently accepted by Cancer Research. In this study, transcription inhibition effects of pyriplatin were investigated in mammalian cells. Pyriplatin inhibits transcription as strongly as bifunctional platinum drugs such as cisplatin and oxaliplatin, and NER plays a major role in removing pyriplatin-DNA lesions. This study provides a rational basis for the design of more potent platinum anticancer drug candidates using a monofunctional DNA-damaging strategy.
